In this webinar, we will present the first demonstration of incorporating T cell isolation upstream of Quantum Flex™ processing, extending automation and process integration from cell selection through expansion.
Starting with Leukopak material collected on the Spectra Optia®, CD3+ T cells were isolated using a negative selection buoyancy-based microbubble separation method. For this autologous process, under 10 million isolated and activated T cells were then directly introduced into the Quantum Flex system, where they underwent lentiviral transduction and subsequent expansion to a therapeutically relevant dose.
Attendees will gain insight into how this integrated, closed-system approach compares with traditional static culture methods and explore data highlighting key process advantages. The workflow is designed to reduce operator intervention, minimize hands-on processing time, and shorten the path from starting material to target cell dose. In
addition, the process demonstrates robust expansion from a relatively small starting cell number while maintaining critical quality attributes throughout manufacturing.
By combining cell isolation, activation, transduction, and expansion into a streamlined workflow, this approach has the potential to simplify manufacturing operations, reduce open handling steps, and support greater process consistency. The presentation will also discuss how integrating buoyant microbubble-based T cell isolation with the Quantum Flex platform may help advance scalable manufacturing strategies for gene-modified T cell therapies.
